                              報告摘要: 

                              腫瘤的發生與演進起源于遺傳和表觀遺傳的異常變化。這些變異導致癌基因的激活、抑癌基因的失活、以及其他分子和信號通路的變化,使正常細胞發生轉化、癌細胞進一步惡化。發現、鑒定腫瘤驅動基因并研究它們的相關信號通路、分子網絡及調節機制對于改善癌癥的檢測、預防與治療發揮著決定作用。利用遺傳、生化、分子細胞生物以及功能檢測的各種手段和細胞、小鼠以及人體組織等各種研究材料,我們多年致力于腫瘤基因的發現和功能、機制方面的研究。先后發現了KAI1(抗癌1號)腫瘤轉移抑制基因、KLF5ZFHX3抑癌基因以及ERRFU50FOXA1WWP1等多個腫瘤基因。我們還對KLF5ZFHX3進行了較深入的功能和機制方面的研究,發現KLF5通過作為TGFβ信號通路的一個關鍵因子實現其抑癌作用、KLF5乙酰化狀態決定KLF5TGFβ發揮抑癌功能還是促癌功能、ZFHX3通過誘導細胞分化來實現其抑癌功能、ZFHX3與激素信號相互作用并調節上皮細胞分化。將基因組和蛋白質組學手段應用到腫瘤材料將導致對腫瘤的各種生物學行為的深入全面的了解,從根本上改變對腫瘤的檢測、預防、藥物開發以及治療方案的制定。 

                              報告人簡介: 

                                Dr. Jintang Dong has expertise in cancer genetics and biology that is helpful for the success of this proposal. Both of his training and independent research have been on these areas. By publishing more than 100 research articles, he has the expertise in multiple areas of cancer biology research, including the discovery and characterization of cancer genes; functional evaluation of cancer genes in various models such as in vitro cultured cells, xenograft tumor models and genetically modified mice; understanding of how cancer genes function in both normal tissues and tumor cells; how genes are transcriptionally regulated; and how cancer genes regulate the balance of cell proliferation and differentiation in epithelial homeostasis. He has also been involved in head and cancer research at Emory, as he has been the Director for the Developmental Research Program in the Emory SPORE in Head and Neck Cancer. He has mentored multiple junior faculty members during his career so far.
